Human enkephalinase (EC 3.4.24.11) is a Zn-containing metalloenzyme that hydrolyzes enkephalins (ENK) at their Gly5-Phe6 peptide bond, which serves as an essential step in the catabolism of these opioid peptides. This report concerns the synthesis of a small library of 20 new chiral model compounds that mimic the backbone of the enkephalins and are being used for initial evaluation of their ability to inhibit or to discriminate against enkephalinase. Each of the 20 compounds has a primary amine functionality at the C-terminus of the molecule and a short side chain alpha-methyl group at position 1. Three of the compounds were prepared in optically active forms by asymmetric synthesis using the chiral auxiliary L-serine ((R,S)-trans-1-phenylethylamine). These three compounds were evaluated for their ability to inhibit enkephalinase by comparing their inhibitory potency to that of the naturally occurring enkephalins (ENK). These studies led to the identification of one compound that has the same inhibitory potency as the natural enkephalins and is thus highly potent and selective. It was also found that there is good correlation between the hydrolysis of the enkephalins and the inhibition of their enzyme. The equilibrium between the partially protected intermediates and the reaction products was found to be the rate-determining step. Finally, the ability of these compounds to discriminate against other zinc enzymes (L-aspartate dehydrogenase and cystathionine gamma-synthase) was also investigated.
